The biggest difference between the front of the D5 and the D4s is the addition of a new programmable button in between the grip and the lens mount.

This is where we start to see some differences. Nikon has replaced the Mode button with the ISO sensitivity control. Not a big deal, unless your muscle memory kicks in. The top of the grip seems to be a steeper angle, and the record button has moved slightly as well.

The only new connection option is the switch from USB 2.0 to USB 3, essential for transferring 4K and the larger files produced by the higher resolution sensor quickly.
