I can't find any analog audio output. I see optical but no jack slot.
Can you share what page of http://downloadcenter.samsung.com/content/UM/201004/20100419101109484/BN68-02589B-00L04_0326.pdf you are connecting to?

Hello everyone,
So I have a Samsung TV, model number: LE32C350 for about 5 years now. Works perfectly and today I decided to buy a 2.1 speaker system. I tried it on my computer and works great, but when i plug it in the jack slot of the TV nothing happens. When I turn the TV Off the sound goes to my speakers but when I turn it on again it goes back to the TV speakers. Can anyone give me some advice on what to do?

That is the model. I plugged the jack in that black slot on page 7 of the manual.
Since you want to output from the TV, then you'll need an output. All I see is that Optical output which you need a receiver or get a soundbar for that.
I wish you could clarify what you mean by slot. The only slot I see is the "common interface slot" but that is for a card and not speakers.
On page 8 of the manual there is a Jack slot for audio out and i tried it there, but doesn't work.
Better put, based on the manual what would I need / have to do to set up my 2.1 speaker system ?

I can see why some may find it confusing as they show output from the devices.
As to your next question, you need a soundbar (with optical support) or a receiver.
Some folk try those Optical to Analog converters.
you may need to tell the tv to use external speakers. go into the tv's settings menu and check.
The speakers are external, analog and apparently plugged into the TV input jacks.
Think about it.
I tried that but after i selected the external speakers the sound went off completely.
