From what it sounds like, you're splitting the signal from one source to two televisions, correct?
If it's not the input setting, the signal may be too weak. The "Check Signal Cable" is basically letting you know that there's either no signal, or the signal isn't strong enough to create a solid connection.
You might try calling the Samsung customer service center in your region to see if additional troubleshooting may be available. If you use a shorter cable or bypass the splitter, are you able to get a signal?

I've got sky and its linked to my Samsung 17inch upstairs. Its not multi-room, its one of the little mouse things which sit on the top of the tv upstairs. Basically whatever channel is on downstairs is whats on upstairs
Anyhow, I've just put the samsung tv on and the screen is blank with a message saying 'check signal cable'
Everything is connected okay and the tv & sky downstairs are working fine.
Any ideas what might be wrong with it, if it can be fixed or is it kaput and I need a new one!
Any help or advice greatly appreciated.
